A Small Column for a Large Volume of Cell Media - The larger the sample volume, the longer it takes to extract pure and native EVs out of it? Not with our SmartSEC-TC. Building on the proven SmartSEC platform that powers our popular SmartSEC Single, SmartSEC HT Plate, and SmartSEC Mini Kit, SmartSEC-TC combines the efficiency and purity of size exclusion chromatography (SEC) with an advanced contaminant-trapping technology in a single 500µl fraction. Compared to both resin-based and bead-based methods, SSEC-TC delivers superior EV isolation results in terms of yield, purity, and overall integrity. The best part is the elimination of multi-fraction collection required with conventional SEC; therefore, delivering higher EV concentration. The kit includes an initial concentration step to reduce large tissue culture media volumes to a manageable size for downstream processing. SmartSEC-TC Achieves Superior EV Purity Compared to Conventional SEC
Figure 2. SSEC-TC delivers higher yields of cleaner EVs compared to multiple pooled fractions obtained from conventional SEC (Company Q). To compare SmartSEC-TC with a competitor’s SEC-based columns, EVs were isolated from 10 mL of HEK293 cell culture media containing 10% exosome-depleted FBS (ExoFBS) using both methods. Western blot and fluorescent nanoparticle tracking analysis showed that SmartSEC-TC produced EVs with comparable particle concentrations but significantly higher EV marker proteins (CD63, CD9) and lower carryover proteins like BSA.

SmartSEC-TC efficiently isolates EVs from serum-free HEK293 cell culture media with high yield and purity, overcoming the challenges of reduced EV yield under serum-free conditions. EVs were isolated from 10 mL of conditioned media using SmartSEC-TC and Company N’s resin-based method. Western blot analysis revealed higher levels of EV marker proteins (CD9, CD81) and minimal BSA carry-over with SmartSEC-TC (A). Additionally, fNTA demonstrated over 6-fold higher particle concentrations and 3-fold greater purity with SmartSEC-TC compared to Company N (B).

EVs Isolated from SmartSEC-TC Contain High Quality RNAs

RNA samples were extracted using SBI’s EVery EV RNA Isolation Kit (Every100B-1) and analyzed with Agilent Bioanalyzer. The data showed excellent size distribution, minimal genomic DNA contamination, and high-quality small RNA enrichment within the 25-200 nt range.
