myBaits Expert Wheat Exome Panel

Enrich for over 200 megabases of high-confidence exons from a wide variety of wheat cultivars

Product Description

  • Developed with the International Wheat Genome Sequencing Consortium
  • 2 million probes targeting >200 megabases of high-confidence exons
  • Compatible with Triticeae, Aegilops, Hordeum, and Secale species, and with any genome ploidy level
  • Pairs with all library preps and sequencing platforms

Acquire deep read coverage at tens to hundreds of megabases of diploid, tetraploid, and hexaploid wheat varieties. Whether for novel variant discovery, detecting introgression, or even routine comprehensive genotyping, the myBaits® Expert Wheat Exome kit reduces sequencing costs by an order of magnitude or more compared to WGS.

Developed in collaboration with the International Wheat Genome Sequencing Consortium (IWGSC) using the Chinese Spring RefSeq v1.0 assembly and v1.1 annotation set, this kit tiles 2 million baits across more than 200 Mbp of target space in hexaploid wheat. The dedicated capture protocol was optimized through trial captures on dozens of hexaploid and tetraploid varieties, offering specificity and sensitivity superior to alternatives in the literature.

myBaits kits can be easily integrated into your own research workflows or paired with our customizable myReads Sequencing or Bioinformatics services for a seamless NGS solution. Our lab services experts have processed thousands of wheat and other cereal samples using the myBaits Expert Wheat Exome v1 probe set and are ready to help with yours.

The Wheat Exome v1 panel is provided as a complete in-solution target capture kit including buffers, blockers, and baits, along with an easy-to-use protocol.

Fig. 1. The Wheat Exome panel can be used on both hexaploid and tetraploid cultivars. Although the panel is designed for all high-confidence exons from the hexaploid Chinese Spring IWGSCv1 reference genome, it has high efficiency for other types of wheat as well. This figure depicts both the predicted vs. observed capturable portions of exomes from hexaploid and tetraploid wheat cultivars. Performance Data

The behavior of this kit has been evaluated across various Triticeae, Aegilops, Secale, and Hordeum species, observing high target specificity and recovery even with distantly related taxa.

The myBaits Expert Wheat Exome panel demonstrates excellent target coverage across a wide range of cultivars. The combination of exonic and non-exonic coverage obtained with the Wheat Exome kit enables it to serve as an effective hexaploid and tetraploid genotyping tool. Even for highly divergent wild emmer wheat (Zavitan), it is easy to achieve high coverage depth at regions of enhanced SNV density.

Reliable and Comprehensive Sequencing Coverage

With only ~18Gbp sequencing effort per sample, the Wheat Exome v1 kit consistently provides an average 34X unique read coverage across >200 Mbp of target space, overlapping >95% of the IWGSC v1.1 high-confidence exon annotations.
