Recommended dilutions WB: 1/1,000 when using colorimetric substrates such as BCIP/NBT and 1/5,000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples. Predicted molecular weight: 43 kDa. This antibody identifies bands at 55 kD and 53 kD when used against the reduced protein as well as activation/breakdown products_recover. The antibody can recognize native or reduced PKARIb. Dilution optimised using Chromogenic detection. Not yet tested in applications. Optimal dilutions/concentrations should be determined by the end user.

Background

The antibody is a rabbit antibody to the regulatory subunit Ib of protein kinase-A, made with a synthetic peptide based on the rat sequence. The antibody binds to PKARIb, but does not cross react with the PKA regulatory subunits (PKARIa, PKARIIa, PKARIIb). The antibody has been peptide-affinity purified, concentrated to 1 mg/ml, with the addition of Sodium Azide as preservative, and 50% glycerol as as cryoprotectant. Protein KinaseA is a critical kinase involved in numerous biological functions. The regulatory subunits of PKA allow a range of modifying agents to influence PKA activity and localization, and are found in different levels in a tissue-specific manner. RP1PKARIb works well for Western Blotting applications, and when used against the reduced protein identifies bands at 55 kD and 53 kD, as well as activation/breakdown products_recover. The antibody can recognize native or reduced PKARIb, and has application for immunoprecipitation, immunohistochemistry, cell sorting and ELISA. RP1PKARIb recognized human, rat, mouse and rabbit PKARIb. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates.
