Background
This serine/threonine protein kinase participates in various signaling pathways involving cytoskeleton regulation, cell motility, cell cycle progression, apoptosis, and proliferation. It's activated by CDC42 and RAC1, leading to autophosphorylation on multiple serine/threonine residues. Full-length PAK2 promotes cell survival and growth, phosphorylating MAPK4, MAPK6, and MAPKAPK5 to influence F-actin polymerization and cell migration. It phosphorylates JUN for EGF-induced proliferation and other substrates like histone H4, BAD, ribosomal protein S6, and MBP. It also functions alongside ARHGEF7 and GIT1 for kinase-independent roles, including spindle orientation during mitosis. In response to apoptotic stimuli, caspase-mediated cleavage generates an active fragment called PAK-2p34, which translocates to the nucleus, triggering apoptosis via the JNK pathway. This caspase-activated PAK2 phosphorylates MKNK1 and inhibits cellular translation.
