Klenow is the large fragment of DNA polymerase I that retains the 5' to 3' DNA polymerase and 3' to 5' exonuclease activities of intact DNA Polymerase I but lacks the 5' to 3' exonuclease activity. It is provided at 5 U/µL.

• Forms blunt ends by removing 3’ overhangs or filling in 5’ overhangs
• Used in second-strand cDNA synthesis
• Generates DNA probes using random primers
