Propagation

PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) are recommended for optimal cell culture. RPMI 1640 Medium (TM503) + 10% Heat-inactivated FBS + 1% Penicillin/Streptomycin Solution (G255), 37.0°C, 5% CO₂.

The Abiraterone Resistant PC-3 Stable Cell Line is a genetically modified variant of the PC-3 human prostate cancer cell line that has been selected for resistance to abiraterone, a potent androgen biosynthesis inhibitor used in the treatment of metastatic prostate cancer. Abiraterone works by blocking CYP17A1, an enzyme involved in androgen production, which is crucial for the growth of prostate cancer cells. However, over time, prostate cancer cells can develop resistance to abiraterone, limiting its therapeutic effectiveness. The cell line was generated by consistently treating PC-3 cells with 10 µM Abiraterone for six months. Resistance was tested by treating cells with 10 µM Abiraterone for at least 24 hours.
