PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) are recommended for optimal cell culture. Dulbecco's Modified Eagle Medium (DMEM), High Glucose (TM500) + 10% FBS(Regular*) + 1X Non-Essential Amino Acids (TM068) + 55 µM 2-Mercaptoethanol (CH045) + 1% Penicillin/Streptomycin Solution (G255), 37.0°C, 5% CO₂. 2.0 µg/ml Puromycin (G264) for selection. This parental SV40T MEF cell line, which features a quadruple knockout of the four distinct eukaryotic translation initiation factor 2 (eIF2α) kinases identified in vertebrates—heme-regulated inhibitor (HRI/EIF2AK1), protein kinase R (PKR/EIF2AK2), PKR-like endoplasmic reticulum kinase (PERK/EIF2AK3), and general control non-derepressible 2 (GCN2/EIF2AK4)—has been modified to overexpress HRI/EIF2AK1. It may be used alongside the wild-type, knockout and/or overexpression cell lines listed below to study the effects of eIF2α kinases in the integrated stress response.
