Propagation
The base medium for this cell line is Prigrow II medium available from ABM (TM002). To make the completed growth medium, add the following components to the base medium: 10 ng/ml recombinant murine IL-3, 10% fetal bovine serum (TM999), and 1% Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
Additional Information
c-Kit or stem cell factor (SCF) receptor is a type III tyrosine kinase receptor that has a major role in the hematopoietic system. The c-Kit wild-type-expressing Ba/F3 cells (with GNNK+ isoform) are ideal in studies in relation to c-Kit function, signaling pathways, and biological activities. The GNNK+ isoform is the outcome of the c-Kit gene alternative splicing sites resulting in different isoforms of the tetrapeptide GNNK. It will proliferate in the presence of the ligand SCF, and activate downstream signal transductions. When compared to GNNK- isoforms (Cat. No. T3041), these cells have a slower rate of SCF degradation as the Src inhibitor SU6656 (SUGEN) exerts a weak effect. Apart from the GNNK+ isoform, GNNK- isoform c-Kit wild-type Stable Ba/F3 cells are also available (Cat. No. T3041).
