Additional Information

This novel murine müller glia cell line (RG17) was spontanously immortalized through serial passaging. RG17 cells were isolated from postnatal day 5 GLAST-CreERT; Rbfl/fl mice. Muller glia (MG) gene and protein expression was verified through qpCR, immunostaining and Wesern blot. Calcium imaging was used to assess purinergic signaling. RG17 cells underwent pharmacological perturbations wit2’-3’-O-(4-benzoylbenzoyl) adenosine 5’-triphosphate (BzATP) and KN-62. Overall, the cell line expresses several MG markers and retains purinergic signaling; RG17 cells retain their ionotropic purinergic receptor P2X7. This cell line is suitable for comparative studies between species that have regenerative capacity and would be useful for the the development of cell-based strategies for retinal degenerative disease treatment.
