Small GTP-binding proteins (or GTPases) are a family of proteins that serve as molecular regulators in signaling transduction pathways.

Rho, Rac and Cdc42, 21 kDa proteins, belong to the family of Rho GTPases regulating a

variety of biological response pathways that include cell motility, cell division, gene transcription, and cell transformation.

Like other small GTPases, Rho, Rac and Cdc42 regulate molecular events by cycling between an inactive GDP-bound form and an active GTP-bound form. In its active (GTP-bound) state, Rho, Rac or Cdc42 bind specifically to the Rhotekin Rho Binding Domain (RBD) resp. to p21-binding domain (PBD) of p21-activated protein kinase (PAK) to control downstream signaling cascades.

Cell Biolabs' Rho/Rac/Cdc42 Activation Assay utilizes Rhotekin RBD Agarose and PAK PBD Agarose beads to selectively isolate and pull-down the active form of Rho, Rac or Cdc42 from purified samples or endogenous lysates. Subsequently, the precipitated GTP-Rho, GTP-Rac or Cdc42 is detected by western blot analysis using an anti-Rho, anti-Rac or Cdc42 antibody.

Cell Biolabs Rho/Rac/Cdc42 Activation Assay Kit provides a simple and fast tool to monitor the activation of Rho, Rac or Cdc42. The kit includes easily identifiable coloured Rhotekin RBD Agarose beads and PAK1 PBD Agarose beads, and a GTPase Immunoblot Positive Control for quick Rho, Rac or Cdc42 identification.

Each kit provides sufficient quantities to perform 10 assays for each small GTPase.
