Glutathione is the major intracellular low-molecular-weight thiol that plays a critical role in
cellular defense against oxidative stress in tissues and cells. Glutathione detection kits
available so far, such as the DTNB-enzyme cycling glutathione assay kit or the
monochlorobimane-based assay kit hardly distinguish between reduced glutathione (GSH;
FW: 307) and oxidized glutathione (GSSG; FW: 612).
This novel Glutathione Assay Kit provides a unique and convenient tool for detecting GSH,
GSSG, and total glutathione individually.

In the assay, OPA (o-phthalaldehyde), which has a very low fluorescence background, reacts
only with GSH (not GSSG), to generate strong fluorescence, so that GSH can be specifically
quantified. By adding a Reducing Agent Mix that converts GSSG to GSH, total glutathione
can then be determined. To measure GSSG, a GSH Quencher is added to specifically quench
GSH, preventing it from reacting with OPA (while GSSG is unaffected). Reducing Agent Mix
is then added to destroy the excess amount of GSH quencher and to convert GSSG to GSH.
Thus, GSSG level can be specifically quantified.
The kit also provides a unique procedure and buffer formula to eliminate protein thiol
interference and to stabilize GSH and GSSG in solutions. The assay is easy to perform and
detects 2-400 ng/ul of GSH, GSSG or total glutathione.
