Donor Vectors for Homologous RecombinationThe PiggyBac HR Vector is designed for use as a donor vector for homologous recombination applications, such as targeted gene editing using TALEN and Cas9 technology. The basic premise of this donor vector is to be able to clone in regions homologous to upstream and downstream of the cut site as to replace (or correct) those regions with a different sequence (e.g. to remove single or multiple bases).
The PiggyBac vector contains MCS sites at the 5´ and 3´ ends of the vector that allow cloning in of homologous sequences at those ends ("HR arms"). Once these regions are cloned in, it can be transfected, along with a suitable TALEN pair, into cells for correction of the DNA sequence. Once homologous recombination has occurred in the cells, the entire construct between the 5´ and 3´ homology regions can be excised out by addition of the Super PiggyBac Transposase (see related links below), leaving only those sequences that are necessary for homologous recombination in a footprint-free manner.
The vector is suitable for use with TALEN- and Cas9- based methods that involve homologous recombination of host genomic sequence, so the necessary TALEN pairs or Cas9 cosntructs for a given gene target must have been designed before.
