Pooled Lentiviral Human Genome-Wide CRISPR sgRNA Library
The Human Genome-Wide CRISPR sgRNA Library developed by our Partner Cellecta is the latest tool for performing loss-of-function screens. The CRISPR/Cas9 system can be used for permanent gene knockout by using a combination of sgRNA and a Cas9 nuclease. Target cells are transduced with Cas9-expressing lentivirus and selected to optimal Cas9 expression levels. The pooled sgRNA library is then transduced into cells so that a majority of the cells receives only one sgRNA construct. Loss-of-function screening followed by Next-Gen Sequencing (NGS) of integrated sgRNA from genomic DNA reveals which genes are involved in the biological process studied.

Cellecta´s Human Genome-Wide CRISPR sgRNA Library is available as a Single Module 80K sgRNA Library with 4 sgRNAs targeting each of 19,000 protein encoding genes or as a 3 Module 150K sgRNA Library with up to 8 sgRNAs per target gene, see below.

Custom libraries comprising a few thousand to over 50,000 sgRNAs specific for your target genes as well as next generation sequencing (NGS) services are also available.


Genome-Wide Human Pooled Lentiviral sgRNA Library (80K)
• 1 single module, covering nearly all protein coding genes (over 19,000 gene targets)
• Each gene is targeted by 4 sgRNAs for a total of 80,000 sgRNAs
• sgRNAs are designed using the latest criteria for effective guides described by Doench et al. Nat Biotechnol 2016 and contain the HEAT design sgRNA structure for more robust screening results
• Red fluorescence marker for sorting of transduced cells and puro selection marker
• Compatible with Illumina HiSeq, GAIIx, and NextSeq NGS systems

Pooled lentiviral-based libraries allow you to assay many thousands of effector constructs simultaneously in one experiment. The Pooled Human Genome-wide guide RNA (gRNA) Library from our partner Cellecta covers nearly all human protein coding genes. Each gene is targeted by 4 sgRNAs for a total of 80,000 sgRNA constructs.
Cellecta’s 80K Genome-Wide CRISPR Knockout Pooled sgRNA Library was designed to be small enough (80,000 constructs) to enable convenient genome-wide “dropout viability” screens with most human cell lines.
The library is provided as either plasmid DNA or pseudoviral particles. The lentivirus-based system ensures efficient delivery of the high complexity, pooled library into a wide range of cell types.


Genome-Wide Modular Human Pooled Lentiviral sgRNA Library (150 K)
• 3 modules, together covering nearly all protein coding genes
• Each gene is targeted by up to 8 sgRNAs for a total of up to 55,000 sgRNAs per module
• Modules can be combined for a single genome-wide screen
HEAT sgRNA design for substantially improved quality of sgRNA libraries and screens
• Red fluorescence marker for sorting of transduced cells and puro selection marker
• Compatible with Illumina HiSeq, GAIIx, and NextSeq NGS systems

The Pooled Human Genome-wide guide RNA (gRNA) Library from our partner Cellecta consists of 3 modules covering nearly all human protein coding genes. Each gene is targeted by 8 sgRNAs for a total of 55,000 sgRNA constructs per module. They are provided as plasmid DNA or pseudoviral particles. For a single genome-wide screen, the library modules can be combined.
The lentivirus-based system ensures efficient delivery of the high complexity, pooled library into a wide range of cell types.
The sgRNA library modules are similar to Cellecta´s Pooled Human Genome-Wide shRNA Library targeting the same sets of genes so researchers can use both CRISPR and RNAi for loss-of-function screens.

For gRNA based knockout screens a two vector approach is crucial - see link below.
Cellecta´s pooled KO libraries are cloned in the vector pRSG16-U6-(sg)-UbiC-RFP-2A-Puro. Pre-transduce your cell line of choice with Cas9 expressing lentivirus, e.g.
Cellecta´s pR-CMV-Cas9-2A-Hygro (for order information see link below) into a small population of cells, then these can be selected strongly to express high levels of the nuclease so that, when the pooled sgRNA are introduced, gene knockout occurs more quickly.
