This packaging plasmid mix is generating especially high lenti titers and can be used for the production of 2nd and 3rd generation lentiviral particles, based on transient co-transfection of a lentiviral siRNA or cDNA construct and plasmids expressing structural proteins (the packaging plasmids) in 293T packaging cells.

The lentiviral siRNA or cDNA construct is also expressing functional virus elements such as Psi, RRE, and cPPT, required for efficient packaging whilst the packaging plasmids encode other necessary structural proteins on separate plasmids, mixed in optimized concentrations.
After co-transfection, the siRNA or cDNA construct is efficiently packaged into VSV-G pseudotyped viral particles which can transduce both dividing and non-dividing cells through VSV-G protein, mediating viral entry through lipid binding and plasma membrane fusion (Burns, 1993).
