The Saliva DNA Isolation Kit is designed for isolating genomic DNA from from up to 250 μl of saliva (or 500 μl of preserved saliva). The saliva sample is collected by spitting or swab and is mixed with the provided Lysis Solution. The resulting saliva sample can be processed immediately, or alternatively can be stored at room temperature for up to 1 month prior to DNA isolation.
Purification is based on spin column chromatography. Under the given conditions only the genomic DNA will bind to the column while most of the contaminating RNA and cellular proteinaceous components are removed in the flowthrough. The purified, eluted saliva DNA is suitable for a number of downstream applications including PCR, Southern Blot analysis, sequencing and microarray analysis. This kit is also compatible with the Norgen Buccal DNA Collection and Preservation Kit and Oragene DNA Saliva Collection Kits.

High Quality and Yield of DNA from Saliva Samples.
Total DNA was isolated from 250 uL of eight different saliva samples using Norgen’s Saliva DNA Isolation Kit (Lanes 1-8). For evaluation, 20 uL of each 100 uL DNA elution was run on a 1.0% agarose gel. Note the high yield and quality of the DNA in all lanes. Lane M: Norgen’s HighRanger 1kb DNA Ladder.

Norgen´s Saliva DNA Isolation Kit was used to isolate DNA from 250 uL samples of saliva mixed with equal amounts of Lysis Solution. From the 100 ul elution, 1 ul, 3 ul, 6 ul and 9 uL of saliva DNA was directly mixed to real-time PCR master mix (total reaction volume 20 uL), and the real-time PCR reaction was performed. 5S rDNA was successfully amplified from the different template volumes without any inhibition, indicating the excellent quality of saliva DNA for down stream applications.

Norgen´s Saliva DNA Isolation Kit was used to isolate DNA from 250 uL samples of saliva mixed with equal amounts of Lysis Solution. Eight samples were used from same donor. From the 100 uL elution, 5 uL of saliva DNA was directly mixed to real-time PCR master mix (total reaction volume 20 uL), and the real-time PCR reaction was performed. GAPDH gene was successfully amplified from the different samples without any inhibition and at the same Ct, indicating the isolation consistency and the excellent quality of saliva DNA for down stream applications.
