Pluripotency Reporter Systems

Pluripotency Response Reporter Constructs
• Monitor and quantitate Oct4 and Sox2 activity
• Lentivector-based reporters with Oct4 or Sox2 response element
• Dual reporting system with both GFP and luciferase
• Optional constitutive EF1-Puro or -Neo selection cassette

The pluripotent state of your stem cells can be verified using Oct4 (CR4) or Sox2 (SRR2) conserved enhancer Pluripotency Response Reporter Constructs. These lentivector-based transcriptional reporters express copGFP and luciferase in response to Oct4 or Sox2 activity. They work for both human and mouse stem cells. Using Oct4 (CR4) or Sox2 (SRR2) Response Reporter Constructs, you can track pluripotency in living cells with GFP and quantitate activities of Oct4 or Sox2 with luciferase.

Oct4 (CR4) or Sox2 (SRR2) conserved enhancer elements are cloned upstream of a minimal CMV promoter driving copGFP and luciferase expression in pluripotent cells.


Pluripotency Promoter Reporter Constructs
• Confirm and enrich for pluripotent stem cells
• Lentivector-based reporters with Oct4 or Nanog promoter
• Choice of fluorescent marker - copGFP or RFP
• Choice of selection marker - Zeocin or thymidine kinase

Pluripotency can be tracked in living cells using the Pluripotency Promoter Reporter Constructs. These lentivector-based constructs express copGFP or RFP under control of a stem cell-specific Oct4 or Nanog promoter. Both human and mouse Oct4 and Nanog Promoter Reporter Constructs are available. They can be applied for live cell imaging, sorting and selection of pluripotent cells.

Oct4 or Nanog promoters drive expression of the fluorescent protein and selection marker in pluripotent cells. Different configurations with copGFP or RFP for sorting, and Zeocin resistance for positive selection or thymidin kinase for negative selection are available.

All Pluripotency Reporter Constructs are available as lentivector plasmids or as pre-packaged lentiviral preparations. Positive and negative transduction control plasmids and pre-packaged viral particles are also provided.
