A Human Induced Pluripotent Stem (iPS) Cell Line is offered with matched source fibroblasts. They enable you to directly compare the differentiated and the induced pluripotent state.
How the iPS Cell Lines are made
Human foreskin fibroblasts (HFFs) are transduced with high-titer iPSC virus to induce pluripotency. The result is high quality iPS cells with no fluorescent markers or drug selection required. The entire process takes up to 6 weeks.
Validation of Pluripotency
The pre-made iPS cell lines are ready for your research and are validated as follows:
Stem cell markers for SSEA-4 (see image below) and Nanog were determined by immunocytochemistry using primary antibodies for SSEA-4 and Nanog followed by fluorescent-labeled secondary Alexa594 antibodies. Detection of Alkaline Phosphatase activity was performed using an AP Detection Kit (see image below). Teratoma assays were performed by subcutaneous injection of 1x10e6 iPS cells into the dorsal flank of SCID mice. Teratomas were visualized after 4 weeks with hematoxylin and eosin staining.
Stem cell marker SSEA-4 was detected by immunocytochemistry.
Alkaline phosphatase activity was confirmed using an AP Detection Kit.
Kit Contents
2 x 10e5 human iPS Cells are delivered together with 50 ml StemPure complete media, as well as with 2 x 10e5 Human Neonatal Foreskin Fibroblast source cells, which are also available in larger quantitities (1 x 10e6) as separate product in 2 formats: From an individual source for generating your own induced pluripotent cells, or pooled for the use as feeder cells.
StemPure Media for growing iPS cells or embryonic stem cells is also available separately in a larger quantity (500 ml).
