TRISOPOR-Protein A consists of recombinant Protein A containing IgG-binding regions (S. aureus) covalently linked to porous glass beads. TRISOPOR is an inert glass matrix devoid of shrinking or swelling. Nanotechnology is employed to create bead pores with a large binding surface leading to the extremely high binding capacity of 45 mg IgG/ml matrix.
TRISOPOR-Protein A can be used for the purification of monoclonal as well as polyclonal antibodies of IgG type from serum, ascites or cell culture.
It is applicable in columns or batch procedures. TRISOPOR-Protein A is provided as a 50% slurry. Prepacked FPLC columns compatible with all common FPLC systems such as ÄKTA, BioCAD and BioSepra are also available.
IgG purification using a prepacked TRISOPOR-Protein A column.
A prepacked TRISOPOR-Protein A column (1 ml) was loaded with 2 ml of human serum diluted 1:1 with 20 mM sodium phosphate buffer pH 7.0 at a flow rate of 0.25 ml/min. After washing, IgG was eluted with 0.2 M Tris/Glycine pH 2.6 at a flow rate of 1.0 ml/min. 0.5 ml fractions were collected and analyzed by agarose gel electrophoresis.
