Cas9 Genome Editing using guide RNAs
The recent discovery of the type II prokaryotic CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) system has provided a revolutionary tool for targeted genome engineering.
The system uses a nuclease, CRISPR-associated (Cas9), that complexes with small RNAs as guides (gRNAs) to cleave DNA in a sequence-specific manner upstream of the protospacer adaptor motif (PAM) in any genomic location. The CRISPR-Cas9 system is comprised of separate guide RNAs known as the crRNA and tracrRNA. These two separate RNAs have been combined into a single RNA to enable site-specific mammalian genome cutting through the design of a short guide RNA.
A series of all-in-one CRISPR-Cas9 cloning vectors with different promoter options is offered. These cloning vectors will express a human codon-optimized Cas9 nuclease as well as any gRNA sequence of your choice linked to the tracrRNA.
TALEN Genome Engeneering Techniques
TAL Effectors (TALE) are proteins expressed and secreted by Xanthomonas bacteria, a plant pathogen. The DNA binding domain contains a highly conserved 33-34 amino acid sequence with the exception of the 12th and 13th amino acids. These two regions contain the highly repeat variable diresidues (RVDs) that demonstrate a correlation with specific nucleotide recognition sequences.
This simple relationship between di-residue amino acid sequence combinations and DNA recognition enables the engineering of highly sequence specific DNA binding domains by selecting a combination of di-residue repeat segments containing the appropriate RVDs.
